Target Information

Signal transducer and activator of transcription 3 (STAT3) functions as both a signal transducer and a transcription activator, mediating cellular responses to interleukins, KITLG/SCF, LEP, and other growth factors. Upon activation, it recruits coactivators, such as NCOA1 or MED1, to the promoter region of target genes. STAT3 plays a crucial role in mouse embryonic development. Research indicates that STAT3 gene knockout results in rapid embryonic degeneration during the early embryonic stages (6.5 to 7.5 days), preventing development to the blastocyst stage and ultimately causing embryonic lethality. This highlights STAT3's essential role in embryonic development. Within mouse mitochondria, STAT3 is involved in regulating the activity of complexes I and II of the electron transport chain (ETC), thereby maintaining cellular homeostasis and energy metabolism. Furthermore, STAT3-deficient mice exhibit mitochondrial dysfunction, characterized by imbalanced regulation of mitochondrial DNA methylation and disrupted imprinted transcription, which adversely impacts both embryonic development and physiological functions in adulthood.
