Rat nephroblastoma overexpressed gene (NOV/CCN3) is a secreted matricellular protein encoded by the Nov gene in Rattus norvegicus, mapped to chromosome 8. It belongs to the CCN protein family, featuring a conserved modular structure including IGF-binding domain, VWC module, TSP1 domain and a C-terminal CT domain. It localizes primarily to extracellular matrix and interstitial fluid, with tissue-specific expression in rat kidney, vascular tissues, brain and bone, with minimal levels in visceral organs. The protein mediates cell-matrix interaction and cell adhesion regulation, with conserved motifs critical for structural stability and ligand binding. Rat NOV shares core functional domains with mammalian orthologs, and has distinct tissue distribution differentiating it from other CCN family members.
