Target Information

Rat myogenin is a 224-residue basic helix-loop-helix transcription factor that binds the consensus E-box motif CANNTG within muscle-specific promoters. The bHLH domain (residues 85-140) mediates DNA contact and obligatory heterodimerization with E12/E47, while an adjacent C-terminal activation region rich in alanine and serine drives transcriptional output. Expressed exclusively in differentiating myoblasts and mature skeletal muscle fibers, the protein localizes to euchromatic nuclei and myonuclei. Antibodies raised against full-length or bHLH-specific peptides detect native, phosphorylated and truncated isoforms with negligible cross-reactivity to MyoD or Myf5, enabling precise immunoblot and immunohistochemical detection in rat skeletal-muscle homogenates and cultured myotubes.
