Target Information
Rat integrin αD (ITGAD) is a 1,145-residue type I transmembrane glycoprotein that pairs exclusively with β2 to form the αDβ2 heterodimer. Its extracellular region contains a seven-bladed β-propeller, an α-β hybrid domain, three calcium-binding thighs, four EGF-like repeats and a novel C-terminal calf domain that stabilizes the bent conformation. The cytoplasmic tail carries an internal GFFKR motif and two threonine-rich inserts that regulate Rap1 binding. Restricted to myeloid lineage, αD is abundant on splenic macrophages, dendritic cells and activated granulocytes. Antibodies raised against the rat-specific calf-2 peptide detect native αDβ2 heterodimers with negligible cross-reactivity to αM, αX or other integrin α chains, enabling selective immunoblot and flow-cytometric detection in rat leukocytes and tissue homogenates.
