Target Information
Mouse proliferating cell nuclear antigen (PCNA) is a 29 kDa toroidal homotrimer whose inter-domain connector loop and C-terminal tail embrace DNA, encircling the helix to tether replicative polymerases and sliding factors. A solvent-exposed 12-aa "KVE" surface loop (aa 164-175) between β-sheets 12 and 13 folds into a β-hairpin unique among mouse DNA clamp proteins, presenting a conformational epitope preserved after nucleotide incorporation and fixation. Antibodies targeting this loop trace native PCNA in S-phase nuclei, replication foci, and repair patches, delivering crisp nuclear puncta and single-band clarity on immunoblots with no cross-reaction to the 9-1-1 clamp or other sliding clamps.
