Target Information

In mice, histamine plays diverse roles, including in immunoregulation, inflammatory response, neurogenesis, and neural function. These effects are primarily mediated through the activation of histamine receptors, such as H1, H2, H3, and H4. Histamine significantly influences the immune system via these receptors. For example, it can modulate the balance between T cell subsets (Th1 and Th2) and affect antibody class switching, like the production of IgE. Mice lacking HDC, which is crucial for synthesizing histamine, show a markedly reduced response to allergic conditions, evidenced by decreased airway hyperresponsiveness, eosinophilia, and lower specific IgE levels. In models of LPS-induced inflammation, histamine can suppress the release of inflammatory factors like IL-1β and HMGB1 and can reverse microglial activation. Histamine is also crucial for mouse behavior and cognitive functions. For instance, H1 receptors located in the brain are linked to epileptic seizures and sleep-wake cycles. Additionally, histamine is involved in the regulation of learning and memory; its release in hippocampal neurons is closely associated with the formation of long-term memory.
