Target Information
The advanced glycosylation end product-specific receptor (RAGE) is a cell surface pattern recognition receptor that detects endogenous stress signals through a wide range of ligands, including advanced glycation end products, S100 proteins, high-mobility group box 1 protein (HMGB1), amyloid beta/APP oligomers, nucleic acids, phospholipids, and glycosaminoglycans. RAGE is linked to various diseases and is crucial in inflammatory responses. For instance, in osteoarthritis, PPARγ (peroxisome proliferator-activated receptor γ) protects chondrocytes by inhibiting RAGE-mediated inflammatory responses. Mice missing RAGE exhibit different pathological responses to certain infections; for example, they have decreased tolerance to Streptococcus pneumoniae infection but experience milder infections with Staphylococcus aureus, while also developing more skin abscesses and experiencing delayed healing.
