Target Information

Tumor necrosis factor receptor superfamily member 9 is a receptor for TNFSF9/4-1BBL. It transmits signals that enhance the survival, cytotoxicity, and mitochondrial activity of CD8+ T-cells, thereby bolstering immunity against viruses and tumors. TNFRSF9 acts as a T cell co-stimulatory factor, activating T cells through interaction with ligands like CD137L, which plays a crucial role in immunomodulation. In mice, TNFRSF9 is primarily expressed on CD8+ T cells and regulatory T cells (Tregs) and is involved in promoting T cell proliferation, preventing apoptosis, and enhancing the secretion of effector factors. Furthermore, TNFRSF9 is crucial in regulating T cell differentiation. For instance, in NOD mice, mutations in the TNFRSF9 gene significantly influence the development of type 1 diabetes (T1D).
