Target Information

GrK, similar to Granzyme A (GzA), induces single-stranded DNA breaks in target cells by cleaving the SET complex in chromatin, leading to cell apoptosis. GrK also has non-cytotoxic roles, such as regulating immune responses and promoting the release of inflammatory factors. Research has demonstrated that GrK can trigger the secretion of proinflammatory cytokines IL-6, IL-8, and MCP-1 by activating the protease-activated receptor-1 (PAR-1). In mice, the GrK gene has been cloned, revealing that the protein it encodes is highly homologous to human Granzyme A. Although GrK is expressed at lower levels than GzA in mouse cytotoxic lymphocytes (CTLs), it retains its function. For instance, in Granzyme A-deficient mice, GrK can partially compensate for the loss of GzA's cytotoxicity.
