Target Information
Sheep transforming growth factor beta 1 (TGF-β1) is a 112-residue mature cytokine released from a 390-amino-acid pro-protein through furin cleavage. The biologically active entity is a 25 kDa disulfide-linked homodimer stabilized by nine intra-chain and one inter-chain cystine, adopting an extended "cystine-knot" fold that presents two symmetrical receptor-binding surfaces. Stored in platelet α-granules and secreted by activated leukocytes, fibroblasts and endothelial cells, the dimer circulates bound to latency-associated peptide and latent TGF-β binding proteins. Antibodies raised against recombinant ovine TGF-β1 detect both the latent complex and mature dimer with negligible cross-reactivity to TGF-β2 or TGF-β3, enabling direct measurement in sheep plasma, tissue extracts and culture supernatants.
