Lysophosphatidic acid (LPA) is a crucial bioactive phospholipid with diverse physiological functions. In the immune system, LPA plays a significant role by converting monocytes into macrophages, a process confirmed in both mice and humans. This conversion occurs through the activation of the Akt/mTOR signaling pathway, with PPARγ as a key transcription factor, highlighting LPA's importance in regulating the innate immune system. Additionally, LPA is involved in mitigating secondary injuries in mice following spinal cord injury via its receptors, such as LPAR2, demonstrating its potential impact on the nervous system. In a high-fat diet-induced obesity model in mice, LPA exhibits notable metabolic effects. It reduces blood glucose tolerance in obese mice by inhibiting insulin secretion and disrupting glucose homeostasis. In terms of gastrointestinal function, LPA influences intestinal smooth muscle contraction in mice by activating voltage-sensitive Ca2+ channels. Moreover, LPA is involved in regulating the immune response by affecting the survival and migration of macrophages, further underscoring its immunological significance.
