Glutathione synthetase facilitates the ATP-dependent conversion of gamma-glutamylcysteine and glycine into glutathione. Glutathione, also known as gamma-glutamylcysteinylglycine (GSH), is the most prevalent intracellular thiol in living aerobic cells, crucial for numerous functions such as protecting cells from oxidative damage, assisting in amino acid transport, detoxifying foreign substances, maintaining protein sulfhydryl groups in their reduced form, and serving as a cofactor for several enzymes. As a vital antioxidant and reductive signaling molecule within mammalian cells, GSH's synthesis plays an essential role in mice. The production of GSH is accomplished through two sequential enzymatic reactions: firstly, γ-glutamylcysteine is catalyzed by γ-glutamyl-cysteine synthetase (GCS), and subsequently, GSH formation is catalyzed by glutathione synthetase (GS). In mice, glutathione synthetase (GS) is critical across various functions, being indispensable for metabolism and antioxidant defense from the embryonic stage to adulthood.
