Rat Alveolar Type II Epithelial Cell Line-RLE-6TN was isolated from 56-day-old male F344 rats by airway perfusion with a streptolysin solution. Expression of the SV40-T antigen was negative by nuclear immunostaining and PCR, indicating that these cells were obtained through spontaneous immortalization. This cell line exhibits characteristics of alveolar type II cells, such as lipid inclusions (identified by Phosphine 3R staining and electron microscopy) and the expression of cytokeratins 8 and 19. The cells do not express alkaline phosphatase activity. Several chemokines expressed by RLE-6TN cells have been reported to be similar to those in primary cultures of alveolar type II cells. They are not tumorigenic in nude mice.
