The mouse ovarian tissue sarcoma cell line M5076 originated from a spontaneous reticulum cell sarcoma found in the ovarian tissue of a C57BL/6 mouse. These cells have been confirmed as macrophage-derived through various tests: they exhibit phagocytic activity, express Fc receptors, show antibody-dependent cellular cytotoxicity, and possess macrophage-like cytotoxic effects against homologous tumor cells without needing LPS activation. Furthermore, M5076 cells lack NK cell activity and demonstrate enzyme activities (lysozyme, non-specific esterase, and phosphatase) comparable to mouse peritoneal macrophages. Their ultrastructural features, including phagocytic vacuoles and a lack of tight junctions, also align with macrophage morphology. Karyotype analysis reveals they are sub-diploid, with over 80% of their chromosomes displaying Robertsonian translocations.
