E2 is the critical host-receptor binding protein of VEEV, forming trimers of E1-E2 heterodimers that project from the viral envelope. It mediates viral entry by specifically interacting with the LDLRAD3 receptor in the brain, facilitating high neurovirulence. As the primary target for neutralizing antibodies, E2 contains major antigenic domains that determine host range and tissue tropism. Its C-terminal cytoplasmic tail also interacts with the viral capsid, playing a vital role in virion assembly and budding from the host cell membrane.
