Pneumolysin is a versatile, pore-forming toxin and a key virulence factor of Streptococcus pneumoniae. It belongs to the cholesterol-dependent cytolysin (CDC) family, binding to host cell membranes and oligomerizing to form large transmembrane pores that cause direct lytic damage. Beyond cell lysis, PLY triggers pro-inflammatory responses, activates the classical complement pathway, and induces apoptosis in host immune and epithelial cells. Its highly conserved nature across pneumococcal serotypes makes it a premier candidate for protein-based vaccines.
