Target Information
In Staphylococcus aureus, WalK is the sensor histidine kinase of the WalKR (YycGF) two-component system, the only such system essential for bacterial viability. Located in the cell membrane, WalK detects extracellular signals through its PAS domain and autophosphorylates to activate its cognate regulator, WalR. It acts as a master controller of peptidoglycan metabolism and cell wall homeostasis. Due to its role in regulating autolysins and antibiotic resistance (e.g., VISA/VRSA phenotypes), it is a high-priority target for novel antimicrobial inhibitors.
