GP4 is a critical minor surface glycoprotein that acts as the primary viral attachment protein for PRRSV. It contains a highly variable neutralizing epitope and is essential for mediating entry into porcine macrophages. GP4 forms a functional heterotrimeric complex with GP2 and GP3, which directly interacts with the host cell receptor CD163. This interaction triggers the conformational changes necessary for membrane fusion and the release of the viral genome into the host cytoplasm.
