Target Information
In mice, Von Willebrand Factor (Vwf) is a critical glycoprotein involved in blood coagulation and platelet adhesion. It plays a key role in mediating platelet-subendothelial interactions and binding to platelets to facilitate their aggregation at sites of endothelial damage. Vwf also acts as a molecular chaperone for factor VIII, protecting it from degradation. Studies show that Vwf-deficient mice exhibit prolonged bleeding times and spontaneous bleeding events, closely mimicking severe human von Willebrand disease. Vwf is regulated by the metalloproteinase ADAMTS13, which cleaves Vwf to reduce its multimer size and binding affinity for collagen and platelets. In disease models, Vwf has been implicated in conditions such as thrombotic thrombocytopenic purpura and acute myeloid leukemia, highlighting its potential as a therapeutic target. Additionally, Vwf has been studied in inflammatory bowel disease and acute cerebral hemorrhage, where it modulates immune responses and vascular permeability.
