Mouse Netrin-1 (Ntn1) is a secreted protein crucial for axon guidance and cell migration during embryonic development. It acts as a chemorepellent or chemoattractant, influencing the formation of neural circuits. In addition to its neural roles, Netrin-1 is involved in vascular patterning in developing organs like the kidney. It also regulates bone remodeling by inhibiting osteoblast differentiation and supporting osteoclast activity. Netrin-1 has anti-apoptotic functions through its receptors, such as DCC and neogenin, which can induce apoptosis in the absence of Netrin-1. In disease contexts, Netrin-1 is implicated in cancer progression, promoting tumor growth and metastasis, and enhancing angiogenesis. It also mitigates acute lung injury by inhibiting TLR4/NF-κB signaling.
