Mouse Inosine Triphosphate Pyrophosphatase (Itpa) is an enzyme that plays a crucial role in maintaining nucleotide pool integrity by hydrolyzing noncanonical purine nucleoside triphosphates such as inosine triphosphate (ITP) and xanthosine triphosphate (XTP) into their monophosphate forms, thereby preventing their incorporation into DNA and RNA. In mice, Itpa is highly expressed in tissues like the testis, brain, and thymus. Itpa deficiency in mice leads to severe developmental anomalies, including growth retardation, cardiomyopathy, and increased chromosomal abnormalities due to the accumulation of noncanonical nucleotides. This enzyme is essential for normal cardiac function, as Itpa knockout mice exhibit structural and functional heart defects.
