Mouse Hexokinase-4 (Gck), also known as glucokinase, is a key enzyme in glucose metabolism, catalyzing the first rate-limiting step of glycolysis by phosphorylating glucose to glucose-6-phosphate (G6P). It acts as a glucose sensor in pancreatic β-cells and the liver, regulating insulin secretion and glycogen synthesis. Gck is essential for maintaining blood glucose homeostasis and is expressed in tissues such as the intestine, hypothalamus, and pituitary gland. Gck plays a central role in diabetes pathogenesis. Mutations in the Gck gene are linked to monogenic diabetes forms like maturity-onset diabetes of the young (MODY2) and permanent neonatal diabetes mellitus (PNDM). In high-glucose environments, Gck undergoes epigenetic modifications, such as promoter hypermethylation, which reduces its expression and contributes to insulin deficiency and diabetes susceptibility in offspring.
