Mouse Glutamate Dehydrogenase 1, Mitochondrial (Glud1) is a key mitochondrial enzyme involved in glutamate metabolism. It catalyzes the reversible oxidative deamination of glutamate to α-ketoglutarate, an essential step in the tricarboxylic acid (TCA) cycle. Glud1 plays a crucial role in maintaining glutamate homeostasis at the tripartite synapse and is expressed in various tissues, including the brain, liver, and muscles. In the central nervous system, Glud1 is primarily expressed in astrocytes, where it helps regulate glutamate levels and neurotransmission. It acts upstream of or within the positive regulation of insulin secretion and is implicated in diseases such as familial hyperinsulinemic hypoglycemia 6, hyperinsulinism, hypoglycemia, and late-onset Parkinson's disease. Glud1 deficiency disrupts glutamate homeostasis in the hippocampus and prefrontal cortex, impairing recognition memory and leading to deficits in visual, spatial, and social novelty recognition abilities. Additionally, Glud1 has been shown to determine muscle stem cell fate by controlling mitochondrial glutamate levels, preventing deleterious mitochondrial glutamate accumulation and inactivation of the malate-aspartate shuttle in proliferating muscle stem cells.

User Note
- Always centrifuge tubes before opening. Avoid mixing by vortexing or pipetting. Reconstitute in 20 mM Tris, 150 mM NaCl (pH8.0) to a concentration of 0.1-1.0 mg/mL. Aliquot the reconstituted solution to minimize freeze-thaw cycles.
