Target Information

Mouse Caspase-14 (Casp14) is a member of the caspase family of cysteine proteases, which are involved in apoptosis and other cellular processes. Casp14 is predominantly expressed in the epidermis and plays a critical role in the terminal differentiation of keratinocytes, particularly in the formation of the stratum corneum and the establishment of the epidermal barrier. It is responsible for the proteolytic processing of profilaggrin into filaggrin and further into free amino acids, which are essential components of the natural moisturizing factors (NMF) that maintain skin hydration. In mouse models, Casp14 has been shown to protect the skin against UVB-induced photodamage and water loss. Casp14-deficient mice exhibit increased sensitivity to UVB radiation, characterized by higher levels of DNA damage and apoptosis in the skin. Additionally, Casp14 is involved in the degradation of other differentiation-associated proteins, contributing to the proper formation of the cornified envelope. Its role in skin barrier function and response to environmental stress makes it a potential target for therapeutic interventions in skin diseases and conditions related to barrier dysfunction.

User Note
