Target Information
Mouse Disintegrin and Metalloproteinase Domain-Containing Protein 8 (Adam8) is a member of the ADAM family of membrane-anchored metalloproteinases. It plays a role in cell adhesion, migration, and signaling, and is implicated in various diseases. Adam8 is expressed in leukocytes and epithelial cells and has been linked to chronic obstructive pulmonary disease (COPD), acute respiratory distress syndrome (ARDS), and cancer. In COPD, Adam8 deficiency exacerbates lung inflammation and emphysema by reducing macrophage apoptosis and increasing oxidative stress. In ARDS, Adam8 signaling drives neutrophil migration and disease severity, with genetic or pharmacologic inhibition of Adam8 reducing lung injury. Adam8 is also involved in cancer progression, promoting metastasis and tumor growth through interactions with signaling pathways like NF-κB and MMP-9. Adam8 is a potential therapeutic target in inflammatory and neoplastic diseases.
