Target Information

Mouse Dihydrolipoyl Dehydrogenase, Mitochondrial (Dld) is a key mitochondrial enzyme involved in the oxidative decarboxylation of α-keto acids. It is a component of the pyruvate dehydrogenase complex (PDC), the 2-oxoglutarate dehydrogenase complex (OGDC), and the glycine cleavage system (GCS), playing a crucial role in energy metabolism and amino acid catabolism. Dld catalyzes the transfer of electrons from dihydrolipoamide to NAD+, facilitating the oxidation of α-keto acids to their corresponding acyl-CoA derivatives. In mouse models, Dld is highly expressed in tissues with high metabolic activity, such as the liver and muscle, where it is essential for maintaining cellular energy homeostasis. Abnormal expression or mutations in Dld can lead to metabolic disorders, including lactic acidosis and maple syrup urine disease. Additionally, Dld has been implicated in the regulation of white adipocyte browning by activating mitochondrial function through the RAS/ERK pathway.
