Target Information
Mouse Apolipoprotein C-III (Apoc3) is a key regulator of lipid metabolism, primarily synthesized in the liver and intestine. It circulates bound to triglyceride-rich lipoproteins (TRLs) such as VLDL and chylomicrons. Apoc3 inhibits lipoprotein lipase (LPL) activity, slowing the hydrolysis of triglycerides and reducing the clearance of TRLs and their remnants by the liver, leading to elevated plasma triglyceride levels. In mouse models, Apoc3 deficiency results in increased LPL activity, hypotriglyceridemia, and protection from postprandial hypertriglyceridemia. Additionally, Apoc3 can induce inflammation by activating the NLRP3 inflammasome in monocytes, although this effect is lost when Apoc3 is bound to lipoproteins. Apoc3 also promotes atherosclerosis by enhancing the binding of atherogenic lipoproteins to glycosaminoglycans in lesions.
