The L1R protein is a highly conserved, myristoylated transmembrane protein located on the envelope of the Intracellular Mature Virion (IMV). It is essential for viral entry, membrane fusion, and the assembly of infectious progeny. L1R contains six conserved cysteine residues that form critical intramolecular disulfide bonds required for its structural integrity. As a major target for neutralizing antibodies, L1R is a primary candidate for subunit vaccine development and the evaluation of cross-protective immunity among orthopoxviruses.
