H3L is a major immunodominant envelope protein located on the membrane of the intracellular mature virus (IMV). It plays a critical role in viral attachment by binding to host cell surface heparan sulfate, facilitating the initial adsorption phase of infection. Due to its high antigenicity, H3L is a primary target for the host's neutralizing antibody response. This makes it a cornerstone antigen for the development of subunit vaccines, therapeutic antibodies, and high-sensitivity serological diagnostic assays for Monkeypox and other orthopoxviruses.
