NS5 is the largest and most conserved protein in the Japanese Encephalitis Virus, functioning as a bifunctional enzyme essential for viral replication. The N-terminal domain possesses methyltransferase (MTase) activity for RNA capping, while the C-terminal domain contains the RNA-dependent RNA polymerase (RdRp) responsible for synthesizing the viral genome. By suppressing host interferon signaling through the inhibition of JAK-STAT pathways, NS5 also serves as a critical antagonist of the innate immune response and a primary target for antiviral drug development.
