Target Information
E1 is a class II viral fusion protein that constitutes the icosahedral scaffold of the EEEV envelope. It primarily mediates pH-dependent membrane fusion between the viral envelope and host endosomal membranes during entry. In the mature virion, E1 exists as an inactive heterodimer with E2; upon endocytosis and acidification, the dimer dissociates, allowing E1 to form a stable homotrimer that inserts its fusion peptide into the host membrane. Its high conservation across alphaviruses makes it a prime target for broad-spectrum neutralizing antibodies and fusion-inhibiting therapeutics.
