NS5B is the essential RNA-dependent RNA polymerase (RdRp) of the Classical Swine Fever Virus, responsible for the replication of the viral positive-sense RNA genome. It features a canonical "right-hand" architecture consisting of fingers, palm, and thumb domains, along with a unique N-terminal domain (NTD) that is critical for its catalytic activity. NS5B initiates RNA synthesis through a de novo mechanism, producing both negative-strand intermediates and progeny positive-strand RNA.
