nsP1 is a multifunctional protein essential for viral RNA capping and membrane anchoring. It possesses both N7-guanine methyltransferase and guanylyltransferase activities, catalyzing the formation of the 5' cap-0 structure to protect viral RNA from host exonucleases. As the sole membrane-associated non-structural protein, nsP1 targets the replication complex to the host plasma membrane via palmitoylation. It oligomerizes into a dodecameric ring (crown-like structure) that anchors viral "spherules," making it a high-priority target for antiviral drug discovery.
