African swine fever virus minor capsid protein p49, also known as pB438L, plays a vital role in the formation and stabilization of the viral capsid's icosahedral structure. Encoded by the B438L gene, p49 is part of the outer capsid and is located near the capsid vertices, being essential for the formation of these vertices. It associates with the membrane, mediating the docking of the penton complex to the inner membrane and recruiting capsomers to form the penton core, which initiates assembly. The protein is also implicated in viral assembly, as it localizes to the viral particle assembly site and facilitates the formation of virions into an icosahedral structure.
