The p14 protein is a crucial structural component of the African Swine Fever Virus (ASFV) core shell. It is generated through the site-specific proteolytic cleavage of the giant polyprotein pp220 (encoded by the CP2475L gene) by the viral cysteine protease S273R. While its primary role is structural-forming the thick protein layer that encloses the viral DNA-containing nucleoid-p14 also possesses specialized functional properties. It is a DNA-binding protein that is actively imported into the host cell nucleus during the early stages of infection. Unlike its "shuttling" counterpart p37, p14 is imported into the nucleus but is not exported back to the cytoplasm, suggesting it may play a dedicated role in anchoring viral DNA or regulatory factors within the nuclear environment to initiate replication.
