Rat zona pellucida glycoprotein 2 (ZP2), a 120-kDa sperm receptor, is a core component of the rat oocyte zona pellucida (ZP)-a extracellular matrix surrounding maturing oocytes in the ovary. It mediates species-specific sperm-oocyte binding and initial fertilization events via interactions with sperm surface proteins. Beyond fertilization, ZP2 contributes to ZP structural integrity, preventing polyspermy and supporting early embryonic development in rats. Critical for rat reproductive success, its dysregulated expression or structural alterations associate with infertility and fertilization failure. As a key mediator of gamete interaction and reproductive competence, rat ZP2 is a critical target for studying reproductive biology and developing contraceptive or fertility-enhancing therapeutics.
