Target Information
Rat transcobalamin II (TC-II) is a 428-residue secreted glycoprotein that binds vitamin B12 via a high-affinity pocket formed by two juxtaposed α/β domains stabilized by a calcium ion and three disulfide bridges. Synthesized primarily in liver and ileal enterocytes, the 43 kDa polypeptide circulates as a 1:1 holo-transporter complex protecting cobalamin from degradation and delivering it to TC-II receptors on dividing cells. Congenital deficiency produces macrocytic anemia through impaired DNA synthesis. Antibodies raised against recombinant rat TC-II recognize both apo- and holo-forms with negligible cross-reactivity to intrinsic factor, haptocorrin, or albumin, enabling direct quantification in rat plasma and tissue extracts.
