Rat tight junction protein 1 (ZO-1, TJP1) is a 220-kDa scaffolding protein predominantly expressed at epithelial and endothelial tight junctions (TJs) of rat tissues (intestine, kidney, blood-brain barrier). It links transmembrane TJ components (occludin, claudins) to the actin cytoskeleton, stabilizing TJ architecture and regulating paracellular permeability. Beyond barrier function, it modulates signal transduction via interactions with kinases and transcription factors, governing rat cell proliferation, differentiation, and inflammatory responses. Critical for maintaining tissue integrity, nutrient absorption, and immune barrier function, its dysregulation associates with inflammatory bowel disease, renal dysfunction, and vascular leakage. As a core TJ scaffolding mediator, rat ZO-1 is a key target for studying barrier pathobiology, inflammatory disorders, and tissue-protective therapeutics.
