Target Information
Rat thioredoxin reductase 1 (Txnrd1) is a 499-residue cytosolic selenoenzyme whose N-terminal FAD-binding domain and C-terminal redox center containing selenocysteine at position 498 catalyze NADPH-dependent reduction of thioredoxin and low-molecular-weight disulfides. The protein functions as a head-to-tail homodimer bridged by an intersubunit disulfide between Cys59 and Sec498, generating a flavin-selenide redox chain. Constitutively expressed in liver, lung and brain, the reductase localizes to cytosol and nucleus. Antibodies raised against full-length or Sec498-proximal peptides detect native, reduced and glutathionylated isoforms with negligible cross-reactivity to Txnrd2 or glutathione reductase, enabling selective immunoblot and immunohistochemical visualization in rat tissue homogenates and sections.
