Target Information
Rat talin 1 is a 2,549-residue cytoskeletal rod built from an N-terminal 47 kDa FERM domain (F1-F3) that binds β-integrin tails, a central helical bundle spanning residues 482-1,650, and a C-terminal 220 kDa I/LWEQ module that anchors F-actin. The elongated monomer (60 nm) dimerizes via a C-terminal coiled coil, generating a two-headed molecule capable of bridging integrins to the cytoskeleton. Constitutively expressed in fibroblasts, platelets, kidney glomeruli and vascular smooth muscle, talin 1 localizes to focal adhesions, costameres and podosomes. Antibodies raised against recombinant rat talin 1 detect native, calpain-cleaved and phosphorylated isoforms with negligible cross-reactivity to talin 2, enabling precise immunoblot and immunofluorescence analysis in rat tissue homogenates and cultured cells.
