Target Information
Rat T-box protein 21 (Tbx21, T-bet) is a 525-residue nuclear transcription factor whose N-terminal T-domain recognizes the consensus 5'-T(G/C)ACAC(T/A)(C/G)-3' via a helix-turn-helix motif anchored by a hydrophobic core. A proline-rich C-terminal tail recruits chromatin remodelers and is targeted by phosphorylation. Restricted to CD4⁺, CD8⁺ T cells, NK cells and dendritic cells, Tbx21 localizes to euchromatin during lineage specification. Antibodies raised against rat-specific T-domain or full-length protein detect native, phosphorylated and truncated isoforms with negligible cross-reactivity to other T-box family members, enabling selective immunoblot, immunohistochemistry and chromatin immunoprecipitation in rat lymphoid tissues and cultured immune cells.
