Target Information
Rat sex-determining region Y-box 18 (SOX18) is a 414-residue transcription factor whose high-mobility-group (HMG) domain (residues 103-181) bends DNA 70° upon binding the consensus motif 5'-AACAAAG-3'. A C-terminal transactivation domain rich in serine and proline undergoes sumoylation and phosphorylation, modulating nuclear import and stability. Expressed selectively in developing vascular endothelium, lymphatic vessels and adult testis, SOX18 localizes to nucleus during vessel specification. Antibodies raised against the HMG or C-terminal epitopes detect native, sumoylated and truncated isoforms with negligible cross-reactivity to other SOX family members, enabling selective immunoblot and immunohistochemistry in rat tissue homogenates and cultured primary endothelial cells.
