Rat O⁶-methylguanine DNA methyltransferase (MGMT) is a 21-kDa DNA repair enzyme ubiquitously expressed in rat tissues, with enriched levels in liver, kidney, and brain. It directly reverses O⁶-methylguanine DNA adducts-formed by alkylating agents-via suicide transfer of the methyl group to its active-site cysteine residue. This mechanism prevents G:C to A:T transversions and genomic instability, safeguarding rat DNA integrity. Critical for cellular resistance to alkylating chemotherapeutics and environmental mutagens, its dysregulation associates with carcinogenesis and chemoresistance. As a key mediator of DNA damage repair and chemotherapeutic response, rat MGMT is a critical target for studying cancer biology, DNA repair mechanisms, and chemosensitization strategies.
