Target Information

Rat nitric-oxide synthase 2 (iNOS) is a 130 kDa homodimeric flavo-heme enzyme whose N-terminal oxygenase and C-terminal reductase domains generate nitric oxide from L-arginine, fueling antimicrobial and inflammatory responses. A solvent-exposed 16-aa "KVE" loop (aa 450-465) between the heme and BH4 pockets folds into a β-hairpin unique among rat NOS isoforms, presenting a conformational epitope preserved after fixation. Affinity-purified antibodies targeting this loop trace native iNOS in activated macrophages, microglia, and hepatocytes, delivering crisp cytoplasmic puncta and single-band clarity on immunoblots with no cross-reaction to nNOS or eNOS.
