Rat laminin γ2 is a 1,203-residue subunit of laminin-332 (formerly laminin-5), distinguished by an extended N-terminal domain containing laminin-type EGF-like repeats, a coiled-coil region mediating trimer assembly with α3 and β3 chains, and a C-terminal G-module that anchors to cell surfaces via syndecans and integrin α3β1. The polypeptide undergoes physiologic cleavage between domains III and IIIa, yielding secreted 105- and 80-kDa fragments. Predominantly expressed in stratified epithelia, kidney tubules, and involuting mammary gland, laminin γ2 localizes to basement membrane zones and interstitial tracks. Polyclonal and monoclonal antibodies raised against recombinant rat laminin γ2 recognize native and cleaved forms with negligible cross-reactivity to laminin γ1 or other laminin subunits, enabling precise immunolocalization and immunoblot detection in tissue homogenates.
