Target Information
Rat indoleamine-2,3-dioxygenase (IDO1) is a 45 kDa cytosolic heme enzyme whose N-terminal catalytic cleft oxidizes tryptophan to kynurenine, starving pathogens and suppressing T-cell proliferation. A solvent-exposed 14-aa "KVE" loop (aa 160-173) between the heme-binding helix and the substrate gate folds into a β-hairpin unique among rat oxidoreductases, presenting a conformational epitope preserved after fixation. Affinity-purified antibodies targeting this loop trace native IDO1 in activated macrophages, placental trophoblasts, and tumor-draining lymph nodes, delivering crisp cytoplasmic puncta and single-band clarity on immunoblots with no cross-reaction to IDO2 or TDO.
