Rat growth-associated protein 43 (GAP-43, neuromodulin) is a 24 kDa membrane-anchored PKC substrate whose N-terminal palmitylation and IQ motif link calmodulin to the cytosolic leaflet, concentrating at axonal growth cones during development and regeneration. A solvent-exposed 14-aa "KVE" loop (aa 45-58) between the basic effector and calmodulin-binding domains folds into a β-turn unique among rat neuronal proteins, presenting a conformational epitope preserved after phosphorylation and fixation. Antibodies targeting this loop trace native GAP-43 in hippocampal mossy fibers, olfactory bulb glomeruli, and sciatic nerve, delivering crisp axonal trails and single-band clarity on immunoblots with no cross-reaction to CAP-23 or MARCKS.
