Target Information
Rat forkhead box protein M1 (FOXM1) is a 748-residue transcription factor built from an N-terminal repressor domain, a central forkhead DNA-binding motif (residues 235-328) that recognises the consensus TAAACA, and a C-terminal transactivation region rich in proline and acidic residues. The protein exists as three splice variants (FOXM1A, B, C) differing at exon Va and VIIa, localising to nucleus during late G1/S and mitosis. Constitutively expressed in thymus, intestine and regenerating liver, FOXM1 undergoes phosphorylation-mediated conformational switching. Antibodies raised against the forkhead or C-terminal epitopes detect native, phosphorylated and truncated isoforms with negligible cross-reactivity to other FOX proteins, enabling selective immunoblot and immunohistochemistry in rat tissue homogenates and cultured cells.
