Rat ectonucleotide pyrophosphatase/phosphodiesterase 2 (ENPP2, autotaxin) is an 863-residue type II transmembrane glycoprotein that hydrolyzes lysophosphatidylcholine into the lipid mediator LPA. The extracellular portion contains two somatomedin-B-like domains, a catalytic phosphodiesterase domain and a nuclease-like lobe that together form a deep lipid-binding pocket around the active-site threonine. A single transmembrane helix anchors the protein to the plasma membrane of neurons, adipocytes and vascular endothelium, while a soluble 100 kDa form circulates after proteolytic shedding. Antibodies raised against the catalytic or SBM domains detect membrane-bound, secreted and deglycosylated isoforms with negligible cross-reactivity to ENPP1 or ENPP3, enabling selective immunoblot and immunohistochemical visualization in rat plasma, tissue homogenates and cultured cells.
