Target Information
Rat delta-like 1 homolog (Dlk1) is a 45 kDa glycosylated transmembrane protein whose six EGF-like repeats extrude from the cell surface to engage Notch receptors and modulate adipogenic versus myogenic fate. A furin-cleavage motif within the juxtamembrane stalk sheds a soluble 30 kDa ectodomain that circulates and competes for the same ligand-binding pocket. The remaining short intracellular tail lacks catalytic residues but clusters with caveolin-1 to fine-tune downstream signals. Antibodies directed against the membrane-proximal EGF-5/6 interface detect both full-length and cleaved forms in brown fat, skeletal muscle, and serum without recognizing classical Notch ligands.
